Iliescu R, Cucchiarelli VE, Yanes LL, Iles JW, Reckelhoff JF. Impact of androgen-induced oxidative stress on hypertension in male SHR. Am J Physiol Regul Integr Comp Physiol 292: R731–R735, 2007. First published July 20, 2006; doi:10.1152/ajpregu.00353.2006.—Men have higher blood pressure than women, and androgens and oxidative stress have been implicated as playing roles in this sexual dimorphism. The spontaneously hypertensive rat (SHR) is an animal model of both androgenand oxidative stress-mediated hypertension. Therefore, the present studies were performed to test the hypothesis that androgens cause hypertension in SHR in part by stimulating superoxide production via NADPH oxidase. Castration of male SHR reduced blood pressure by 15% and attenuated both basal and NADPH-stimulated superoxide production in kidney cortical homogenates. Expression of p47 and gp91 but not p22 subunits of NADPH oxidase were significantly lower in kidney cortex from castrated males compared with intact males. Moreover, inhibition of NADPH oxidase with apocynin caused 15 mmHg reduction in blood pressure and reduced basal and NADPH-stimulated superoxide production in intact male SHR, but had no effect on blood pressure or superoxide production in castrated males. These data support the hypothesis that androgens cause oxidative stress and thereby increase blood pressure in male SHR via an NADPH oxidase-dependent mechanism.

متن کاملOxidative stress contributes to sex differences in angiotensin II-mediated hypertension in spontaneously hypertensive rats.
NADPH oxidase has been implicated in ANG II-induced oxidative stress and hypertension in males; however, the contribution of oxidative stress to ANG II hypertension in females is unknown. In the present study, we tested the hypothesis that greater antioxidant capacity in female spontaneously hypertensive rats (SHR) blunts ANG II-induced oxidative stress and hypertension relative to males. Whole...
متن کاملIn Focus CALL FOR PAPERS Sex Differences in Renal and Cardiovascular Function: Physiology and Pathophysiology Physiological and molecular mechanisms governing sexual dimorphism of kidney, cardiac, and vascular function
IT IS WELL ESTABLISHED THAT cardiovascular disease is a leading cause of death in developed countries, but it is less well known that more women than men die of cardiovascular disease, although these deaths are delayed by 10 yr (7, 17). It is becoming evident that there is considerable sexual dimorphism in the pathogenesis of cardiovascular disease. Thus findings from past studies, mostly in ma...
متن کاملSex and sex hormones influence the development of albuminuria and renal macrophage infiltration in spontaneously hypertensive rats.
There is a sex difference in hypertensive renal injury, with men experiencing greater severity and a more rapid progression of renal disease than women; however, the molecular mechanisms protecting against renal injury in women are unknown. The goal of this study was to determine whether sex hormones modulate blood pressure and the progression of albuminuria during the developmental phase of hy...
